A chromatographic method for the detection of tryptophan metabolites.

چکیده
During the course of investigations involving the action of tissue slices on tryptophan, the aqueous medium in which n-kynurenine sulfate had been incubated with kidney slices was found to have acquired a marked bluish violet fluorescence. Subsequent investigation showed that several known metabolites of tryptophan fluoresce when suitably exposed to ultraviolet light. Advantage has been taken of this behavior and of separation by paper chromatography to identify a number of the products formed when tryptophan and certain of its metabolites are incubated with slices of rat liver or rat kidney. This communication describes our observations.

Rapid Isocratic Liquid Chromatographic Separation and Quantification of Tryptophan and Six kynurenine Metabolites in Biological Samples with Ultraviolet and Fluorimetric Detection
A simple, rapid isocratic liquid chromatographic procedure with ultraviolet and fluorimetric detection is described for the separation and quantification of L-tryptophan (Trp) and six of its kynurenine metabolites (kynurenine, 3-hydroxykynurenine, and 3-hydroxyanthranilic, kynurenic, xanthurenic and anthranilic acids). Using the Perkin Elmer LC 200 system, a reverse phase Synergi 4 μ fusion-RP8...

An agar well diffusion bioassay method for determination of clarithromycin in human plasma, using Micrococcus Luteus ATCC 9341 as the assay organism, was compared with a selective high-performance liquid chromatographic (HPLC) method with UV detection. Spiked plasma was used to prepare standard and control samples for both methods. The results of the bioassay analyses with spiked plasma samples...

متن کاملComplementary use of amperometric and spectrophotometric detection for concurrent monitoring of serum tryptophan metabolites by reversed-phase liquid chromatography.
We describe an improved reversed-phase liquid-chromatographic method for concurrent measurement of tryptophan and eight of its metabolites in serum. These were separated by gradient elution and detected both by ultraviolet absorption at 254 nm and amperometrically at an oxidation potential of +0.700 V vs Ag/Cl. Detection limits for the electrochemically-detected compounds were between 100 pg an...
متن کاملChromatographic analysis of tryptophan metabolites
The kynurenine pathway generates multiple tryptophan metabolites called collectively kynurenines and leads to formation of the enzyme cofactor nicotinamide adenine dinucleotide. The first step in this pathway is tryptophan degradation, initiated by the rate-limiting enzymes indoleamine 2,3-dioxygenase, or tryptophan 2,3-dioxygenase, depending on the tissue. The balanced kynurenine metabolism, w...
